Nmda Infusion

NMDA infusion is the controlled administration of N-methyl-D-aspartate, an agonist that activates NMDA-type glutamate receptors to investigate excitatory signaling in the nervous system. After binding to these ionotropic receptors, NMDA promotes channel opening, allowing sodium and calcium ions to enter neurons and producing membrane depolarization; sustained activation can lead to excitotoxic stress and neuronal injury. In neuroscience research, NMDA infusion helps model synaptic plasticity, seizure-related activity, neurodegeneration, and excitotoxicity under defined experimental conditions. These models support analysis of neuronal vulnerability, receptor function, and potential neuroprotective strategies, while informing studies of learning, memory, and disease mechanisms.

Here, we describe the use of a genetically directed alkaline phosphatase (AP) labeling method to investigate the anatomical features and phenotypes of mouse retinas under NMDA-induced excitotoxicity. In conjunction with several retinal ganglion cell (RGC) subtype-specific reporter lines, we uncover novel phenotypes in dying RGCs throughout the degenerative process.
