Oligodendrocyte Precursor Cells

Oligodendrocyte precursor cells (OPCs) are proliferative glial cells in the central nervous system that generate oligodendrocytes, the cells responsible for myelinating axons. They respond to developmental and local signals by dividing, migrating, and differentiating, after which mature oligodendrocytes extend processes that wrap axons with myelin to support rapid electrical conduction. OPCs remain widely distributed in the adult brain and spinal cord, where they can participate in ongoing myelin maintenance and repair following injury or disease. Studying their regulation helps neuroscience researchers understand neural development, white matter disorders, remyelination, and potential strategies for restoring circuit function.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.

The video demonstrates a protocol for differentiating oligodendrocyte precursor cells (OPCs) into oligodendrocytes and inducing myelination in a co-culture with dorsal root ganglion (DRG) neurons. The OPCs are transduced with lentiviral vectors encoding a signaling protein that regulates myelination. The transduced OPCs are co-cultured with DRG neurons to assess the differentiation into oligodendrocytes and the signaling protein-induced myelination.

This article describes methods to derive enriched populations of murine oligodendrocyte precursor cells (OPCs) in primary culture, which differentiate to produce mature oligodendrocytes (OLs). In addition, this report describes techniques to produce murine myelinating co-cultures by seeding mouse OPCs onto a neurite bed of mouse dorsal root ganglion neurons (DRGNs).
