Pre-stimulus Activity

Pre-stimulus activity is the ongoing neural activity that occurs before an external sensory event, providing a baseline state that can influence how the brain processes incoming information. It arises from spontaneous neuronal firing, synaptic input, and rhythmic fluctuations in membrane excitability, which can alter the likelihood and strength of stimulus-evoked responses. In neuroscience, researchers measure these patterns with methods such as electroencephalography, functional imaging, and intracranial recordings to examine links between brain state, attention, perception, and behavioral variability. Studying pre-stimulus activity helps explain why identical stimuli can produce different neural and perceptual outcomes across trials and individuals.

Inducing Axonal Varicosities with a Micromechanical Stimulus on Neurons

This video demonstrates the induction of axonal varicosities by applying micromechanical stimulus on neurons. The system comprises a micropipette attached to a syringe containing buffer via tubing. Using a micromanipulator, the micropipette is positioned above the neurons. Fluid pressure is then applied to induce varicosities in axons.

An In Vitro Assay for Studying the Cytotoxicity of Pre-Activated CD8+ T Cells against Cancer Cells

This video showcases an apoptosis assay in cancer cells induced by pre-activated effector T cells. By co-culturing these cells with cancer cells marked with fluorescent nuclei, a caspase-cleavable fluorogenic substrate is added. Apoptotic cancer cells will display a different color in their nuclei, leading to dual fluorescence. This dual fluorescence is indicative of cancer cells undergoing apoptosis.

Steady-state, Pre-steady-state, and Single-turnover Kinetic Measurement for DNA Glycosylase Activity

Time courses for the glycosylase activity of 8-oxoguanine DNA glycosylase are biphasic exhibiting a burst of product formation and a linear steady-state phase. Utilizing quench-flow techniques, the burst and the steady-state rates can be measured, which correspond to excision of 8-oxoguanine and release of the glycosylase from the product DNA, respectively.
