Retinal Quantification

Retinal quantification is the measurement and numerical analysis of retinal structure, cells, or signals to characterize visual-system organization and change. In neuroscience, it typically combines microscopy or imaging with image processing methods that segment retinal layers or cell populations, extract features such as thickness, density, area, or fluorescence intensity, and compare these measurements across samples or conditions. Quantitative analysis can reveal developmental patterns, neuronal loss, synaptic or vascular alterations, and responses to injury or treatment. By converting complex retinal images into reproducible measurements, the approach supports investigations of neural circuits, disease mechanisms, and therapeutic outcomes.

Source: Lani-Louzada, R., et al. Full-Circle Cauterization of Limbal Vascular Plexus for Surgically Induced Glaucoma in Rodents. J. Vis. Exp. (2022).This video demonstrates the quantification of retinal ganglion cells from chemically fixed rat retinas subjected to full-circle cauterization of the limbal vascular plexus. The process includes permeabilization, antibody staining, and nuclear counterstaining, followed by visualization under an epifluorescence microscope to assess the impact of...

(2020).This video demonstrates imaging and quantification of axonal regeneration in axotomized rat retinal ganglion neurons (RGNs) co-cultured with immortalized human olfactory ensheathing glia cells. RGNs are immunolabeled, with red somas and green axons. Images were captured using an epifluorescence microscope,...

We demonstrate a method to label the walls of the retinal vasculature and adherent leukocytes. These adherent leukocytes can then be counted under a fluorescence microscope as a parameter of inflammation or the response of that inflammation to therapies.
