Rpe Monolayer Formation

RPE monolayer formation is the establishment of a continuous, polarized single-cell layer of retinal pigment epithelial cells that supports retinal structure and function. The process depends on cell adhesion, proliferation, and maturation until neighboring cells form tightly connected junctions with distinct apical and basal surfaces. A stable monolayer develops epithelial barrier properties, including selective transport and regulated interaction with photoreceptors and the underlying tissue. In neuroscience, RPE monolayers provide experimental models for studying the blood-retina barrier, retinal development, degeneration, inflammation, and responses to potential therapies, helping researchers evaluate how altered epithelial function contributes to visual disease.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
