Phospholipase C Pathway

The phospholipase C pathway is a cell-signaling system that converts extracellular stimuli into intracellular responses, making it important in pharmacology and therapeutic drug action. Activation of G protein-coupled receptors or receptor tyrosine kinases stimulates phospholipase C, which cleaves membrane phosphatidylinositol 4,5-bisphosphate into diacylglycerol and inositol trisphosphate; these messengers activate protein kinase C and release calcium from intracellular stores. The pathway regulates processes including smooth-muscle contraction, secretion, metabolism, and gene expression. Understanding its components and downstream effects helps researchers explain drug mechanisms, identify therapeutic targets, and predict adverse responses caused by altered receptor or signaling activity.

In this video, we demonstrate an assay to establish the presence of phospholipase in the crude venom extract from Anthopleura dowii—a marine sea anemone. The assay utilizes an agarose gel diffusion technique, in which the enzyme phospholipase diffuses and breaks down phospholipids incorporated in the solid media in a radial manner.

To test the inhibitory effects of pharmacologic agents on phospholipase C (PLC) in different regions of the honeybee brain, we present a biochemical assay to measure PLC activity in those regions. This assay could be useful for comparing PLC activity among tissues, as well as among bees exhibiting different behaviors.
