Aortic Valve Tissue

Aortic valve tissue is the flexible, fibrous material forming the three leaflets that regulate blood flow from the heart’s left ventricle into the aorta. Its layered structure, rich in collagen and elastin, provides anisotropic mechanical behavior, allowing the leaflets to withstand pressure, open with ventricular contraction, and coapt to prevent backflow during relaxation. In physics and biomechanics, researchers study its stress, strain, elasticity, and fluid-structure interactions to understand valve function and disease. These measurements support models of cardiovascular flow and guide the design, testing, and long-term evaluation of prosthetic and tissue-engineered heart valves.

Full-root aortic valve replacement by stentless aortic xenograft is a viable option in patients with small aortic roots. We describe, a technique for the full-root implantation of stentless aortic xenografts, with emphasis on the management of the proximal suture line and coronary anastomoses, and discuss its limitations and alternative options.

This protocol describes the isolation, culture, and calcification of rat-derived valve interstitial cells, a highly physiological in vitro model of calcific aortic valve disease (CAVD). Exploitation of this rat model facilitates CAVD research in exploring the cell and molecular mechanisms that underlie this complex pathological process.

A cyclic pressure bioreactor capable of subjecting heart valve tissue to physiological and pathological pressure conditions has been designed. A LabVIEW program allows users to control pressure magnitude, amplitude and frequency. This device can be used to study the mechanobiology of heart valve tissue or isolated cells.

Calcification Assay to Measure Calcium Concentration in Aortic Valve Interstitial Cells

This video demonstrates an assay technique for assessing the calcification of the aortic valve interstitial cells in vitro. The assay helps to quantify the calcium concentration using the arsenazo III reagent, a calcium-sensitive dye that reacts with calcium ions to form a purple-colored calcium-arsenazo complex.
