Novel Situations

Novel situations are unfamiliar events, environments, or social contexts that require people to interpret new information and adapt their behavior. Because existing knowledge or expectations may not fully apply, individuals attend to distinctive cues, appraise uncertainty and demands, and use learning, memory, and problem-solving processes to guide responses; these conditions can also activate physiological and emotional stress responses. In psychology, studying novel situations helps researchers examine cognitive flexibility, decision-making, social behavior, and adaptation across development and everyday life. Findings can inform theories of learning and resilience, as well as approaches to education, clinical assessment, and support for effective coping with change.

High-throughput Saccharification Assay for Lignocellulosic Materials

A simple, rapid method for determining the saccharification potential of large numbers of plant biomass samples is described. The automated platform for this analysis involves the preparation of the plant biomass for analysis in 96 well plates and the subsequent performance of pretreatment, hydrolysis and quantification of the sugars released.

We describe a method to quantify the aggregation of misfolded proteins. Our protocol details lentiviral induced stable cell line generation, automated confocal imaging, and image analysis of protein aggregates. As an illustrative application, we studied the effect of small molecules in promoting SOD1 aggregation in a time- and dose-dependent manner.
