Structural Protein Function

As proteínas estruturais são uma classe de proteínas que suportam a forma e movimento das células. Fora da célula, estas proteínas estão presentes em partes estruturais do corpo, tais como ossos, cartilagens e pele. O colágeno, a proteína mais abundante em mamíferos, é uma proteína estrutural encontrada em todo o corpo e a matriz extracelular.Esta proteína se dobra em uma tripla hélice que polimeriza em fibrilas alongadas, que então, se juntam em fibras maiores. Entre suas muitas funções estruturais, contribui para a forma da célula e do tecido. O citoesqueleto é predominantemente composto de proteínas estruturais e forma o quadro básico de uma célula.Contém três tipos principais de filamentos, microfilamentos, filamentos intermediários, e microtúbulos. A actina é uma proteína estrutural que compõe os microfilamentos. Esta proteína polimeriza-se em filamentos e contribui para a forma, organização, movimento e divisão da célula.Filamentos intermediários podem ser compostos de diferentes proteínas, dependendo do tipo de célula. E mutações nos genes que codificam para estas proteínas podem resultar em várias doenças, incluindo anomalias esqueléticas e cardiovasculares.

As proteínas estruturais são uma categoria de proteínas responsáveis por funções que vão desde a forma e movimento das células até o fornecimento de suporte a estruturas importantes, como ossos, cartilagens, cabelos e músculos. Este grupo inclui proteínas como colágeno, actina, miosina e queratina. O colágeno, a proteína mais abundante nos mamíferos, é encontrado em todo o corpo. No tecido conjuntivo, como pele, ligamentos e tendões, proporciona resistência à tração e elasticidade. Nos ossos e dentes, mineraliza-se para formar tecidos duros e contribui para a sua capacidade de suporte de carga. Além do suporte estrutural, o colágeno também pode interagir com receptores de superfície celular e outras moléculas intermediárias para regular processos celulares, como crescimento e migração, que envolvem mudanças na forma das células e dos tecidos. As proteínas estruturais formam a estrutura básica do citoesqueleto celular. O citoesqueleto é composto por três tipos de filamentos, microfilamentos, filamentos intermediários e microtúbulos, e cada um é composto por diferentes proteínas estruturais. O microfilamento é formado quando a actina se autopolimeriza em longas estruturas repetitivas. Esses filamentos de actina contribuem para a forma e organização celular; além disso, os microfilamentos também podem contribuir para o movimento e divisão celular, quando atuam em conjunto com a miosina. A composição dos filamentos intermediários varia de acordo com o tipo de célula. Existem cerca de 70 genes diferentes que codificam vários filamentos intermediários. Os filamentos intermediários nas células epiteliais contêm queratina, os neurônios periféricos contêm periferina e o sarcômero nas células musculares contém desmina. Por exemplo, uma mutação num gene que codifica o colagénio pode resultar numa condição conhecida como osteogénese imperfeita, que é caracterizada por ossos fracos e deformidades nos tecidos conjuntivos. Diferentes mutações em um gene do colágeno podem resultar na síndrome de Alport, caracterizada por problemas em órgãos como rins, olhos e ouvidos.

Structural proteins give cells and tissues shape, support, and strength. They help build bones, cartilage, hair, and muscles. Important examples include collagen, actin, myosin, and keratin.

Collagen is the most abundant protein in mammals. It is found throughout the body. In skin, ligaments, and tendons, collagen adds tensile strength and elasticity. In bones and teeth, it helps form hard, load-bearing tissue through mineralization.

Collagen also does more than provide support. It can interact with cell surface receptors and other molecules to help control cellular processes. These processes include growth and migration, which involve changes in cell and tissue shape.

Structural proteins also form the cell cytoskeleton, which is the internal framework of the cell. The cytoskeleton contains three kinds of filaments: microfilaments, intermediate filaments, and microtubules. Each filament type is built from different structural proteins.

Microfilaments form when actin self-polymerizes into long, repeating strands. These actin filaments help maintain cell shape and organization. They also work with myosin to support cell movement and cell division.

Intermediate filaments vary by cell type. Around 70 different genes code for these proteins. Epithelial cells contain keratin, peripheral neurons contain peripherin, and muscle cell sarcomeres contain desmin. Their main role is to reinforce cells and organize them into tissues.

Microtubules are made of tubulin proteins. Tubulins self-assemble into structures that help organize the cytoplasm and position organelles. Microtubules are also essential for mitosis and cell division. Because structural proteins are so widespread, mutations in their genes can cause serious effects. For example, collagen gene mutations can lead to osteogenesis imperfecta, which causes weak bones and connective tissue deformities, or Alport syndrome, which affects the kidneys, eyes, and ears.

Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
