Allosteric Proteins-ATCase

As proteínas que contêm sítios de ligação ligados são chamadas de proteínas alostéricas e podem ser de vários tipos, como enzimas, receptores, proteínas estruturais e proteínas motoras. 
Por exemplo, a aspartato transcarbamoilase, também conhecida como ATCase, é uma grande enzima contendo seis subunidades catalíticas, cada uma com um sítio de ligação ao substrato. Ele também possui seis subunidades regulatórias adicionais, cada uma com um local que pode se ligar a pirimidinas e purinas, os blocos de construção do DNA e do RNA.
A enzima catalisa uma etapa essencial na síntese de pirimidina, a condensação de aspartato e fosfato de carbamoil para produzir aspartato de carbamoil. A taxa dessa reação aumenta de maneira dependente da concentração.
A ligação do ATP, uma purina, à enzima ativa a enzima. Considerando que, a ligação simultânea de UTP e CTP, pirimidinas, resulta em uma inibição de 95% da atividade enzimática. 
Esse mecanismo regulatório é essencial para manter a quantidade certa de pirimidinas em relação às purinas na célula.

Por exemplo, a atividade enzimática é frequentemente regulada através de um mecanismo de feedback onde o produto final do processo bioquímico serve como inibidor. Aspartato transcarbamoilase (ATCase) é uma enzima citosólica que catalisa a condensação de L-aspartato e carbamoil fosfato em N-carbamoil-L-aspartato. Esta reação é a primeira etapa na biossíntese da pirimidina. UTP e CTP, os produtos finais da via de síntese da pirimidina, inibem a atividade da ATCase, a enzima que catalisa a primeira etapa essencial desta via. A ligação de UTP e CTP à enzima regula negativamente o sítio catalítico ligado quando a concentração de pirimidinas é alta, em relação à concentração de purinas na célula. Este fenômeno é conhecido como inibição por feedback e é essencial para manter as quantidades certas de metabólitos em um organismo. ATCase faz parte do complexo multienzimático CAD, parte da via de biossíntese da pirimidina, junto com a carbamoil fosfato sintetase II e a diidroorotase. As pirimidinas são essenciais para a síntese de DNA durante a divisão celular, portanto, a inibição da atividade da ATCase retarda o crescimento do tumor no câncer.

ATCase is a key enzyme in pyrimidine synthesis. It is a cytosolic enzyme that catalyzes the condensation of L-aspartate and carbamoyl phosphate to form N-carbamoyl-L-aspartate. This reaction starts the pathway that makes pyrimidines.

ATCase is regulated by feedback inhibition. The end products UTP and CTP bind to the enzyme and reduce its activity when pyrimidine levels are high compared with purines in the cell. This binding at one site affects the linked catalytic site, which helps keep metabolite levels balanced.

ATCase also works as part of the CAD multi-enzyme complex. CAD includes carbamoyl phosphate synthetase II and dihydroorotase in the pyrimidine biosynthesis pathway. Because pyrimidines are needed for DNA synthesis during cell division, blocking ATCase activity can slow tumor growth in cancer.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
