Antibiotic-supplemented Media

Antibiotic-supplemented media are laboratory growth media containing antimicrobial agents that reduce unwanted microbial contamination during the culture of cells, tissues, or microorganisms. These compounds act through defined targets, such as bacterial ribosomes or cell-wall synthesis pathways, so the selected antibiotic and concentration determine which contaminants are inhibited while minimizing effects on the intended culture. In biology, such media support more consistent cell culture, cloning, and experimental workflows, but they do not replace aseptic technique and may alter cellular behavior or select for resistant contaminants, making appropriate controls and antibiotic-free validation important.

(2018)This video demonstrates the phage-mediated transfer of donor DNA carrying an excisable antibiotic resistance cassette into Escherichia coli. The integrated cassette replaces the target gene via homologous recombination, resulting in a stable gene deletion.

Antibiotic Dereplication Using the Antibiotic Resistance Platform

We describe a platform that utilizes a library of isogenic antibiotic resistant Escherichia coli for the dereplication of antibiotics. The identity of an antibiotic produced by bacteria or fungi can be deduced by the growth of E. coli expressing its respective resistance gene. This platform is economically effective and time-efficient.
