Golgi-resident Membrane Proteins

Golgi-resident membrane proteins are integral membrane proteins maintained within the Golgi apparatus, where they organize and regulate essential steps in intracellular trafficking and cargo processing. Their localization depends on features such as transmembrane-domain properties, cytosolic sorting signals, and retrieval pathways that return escaped proteins from later compartments to the Golgi. These proteins function as glycosyltransferases, transport regulators, and processing enzymes, establishing the sequential environment needed to modify proteins and lipids as they pass through Golgi cisternae. Studying their targeting and retention mechanisms clarifies organelle organization, secretion, membrane biogenesis, and the cellular defects associated with disrupted trafficking.

The precise localization of Golgi residents is essential for understanding the cellular functions of the Golgi. However, conventional optical microscopy is unable to resolve the sub-Golgi structure. Here we describe the protocol for a conventional microscopy based super-resolution method to quantitatively determine the sub-Golgi localization of a protein.
