Dendritic Cell Generation

Dendritic cell generation is the production and differentiation of dendritic cells, specialized antigen-presenting cells that connect innate sensing with adaptive immune responses. In laboratory systems, hematopoietic progenitors or blood monocytes are cultured with cytokines such as granulocyte-macrophage colony-stimulating factor and interleukin-4, then exposed to microbial or inflammatory signals that promote maturation, increased major histocompatibility complex expression, and costimulatory activity. Generated dendritic cells enable researchers to study antigen uptake, processing, and T-cell activation during infection. They also support investigations of vaccine design, immune regulation, cancer immunotherapy, and host-pathogen interactions.

The incubation of monocytes with growth factors and cytokines leads to the production of immature DCs. In response to immune regulatory molecules, immature DCs differentiate into tolerogenic and mature DCs.

Immature dendritic cells can be selectively differentiated into tolerogenic or mature dendritic cells to regulate the balance between immunity and tolerance. This work presents a means to generate from immature monocyte derived dendritic cells (moDCs), in vitro tolerogenic and mature moDCs that differ in metabolic phenotypes.

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.

Evaluated in syngeneic immunocompetent hosts, cancer stem cell (CSC) based dendritic cell (DC) vaccine demonstrated significantly higher antitumor immunity than traditional DC vaccines pulsed with heterogeneous bulk tumor cells.
