Fluorescent Bead Opsonization

Fluorescent bead opsonization is an immunological assay that coats fluorescent microspheres with opsonins, molecules that mark particles for recognition and uptake by phagocytes. During the assay, beads are incubated with serum or purified antibodies and complement, allowing these proteins to bind their surfaces; immune cells then capture the opsonized beads through Fc and complement receptors, producing a measurable fluorescence signal. Researchers use microscopy or flow cytometry to quantify binding and phagocytosis, compare serum or antibody activity, and investigate host defense against infection. The method provides a controllable model for studying particle recognition, complement function, and cellular uptake.

This video demonstrates an assay to monitor independent macrophage-mediated red blood cell (RBC) phagocytosis events. Mouse immunoglobulin G (IgG)-opsonized human RBCs are incubated with mouse macrophages and observed under a confocal microscope. The RBCs are engulfed by macrophages via a phagocytic cup formation and targeted for lysosomal degradation.

This video demonstrates an opsono-adherence assay for examining the interaction between opsonized bacteria and immune cells. Fluorescently labeled bacteria, treated with test serum containing anti-capsule antibodies, are incubated with macrophages. After staining the macrophages with fluorescence, the attached bacteria on the macrophage surface are observed under a microscope for visualization.
