Luminol

Luminol is a chemiluminescent reagent that produces visible light during oxidation, making it useful for detecting chemical and enzymatic activity. In an alkaline solution, hydrogen peroxide oxidizes luminol, while catalysts such as heme-containing peroxidases or metal ions promote formation of an excited 3-aminophthalate intermediate, which releases energy as blue light. In immunology and infection research, luminol-based chemiluminescence supports sensitive measurement of peroxidase activity in immunoassays, analysis of reactive oxygen production by immune cells, and detection workflows linked to microbial or host responses.

We describe a non-invasive imaging method for distinguishing inflammatory stages. Systemic delivery of luminol reveals areas of acute inflammation dependent upon MPO activity in neutrophils. In contrast, injection of lucigenin allows for visualization of chronic inflammation dependent upon Phox activity in macrophages.
