Mdck A549 Cells

MDCK and A549 cells are established epithelial cell lines used to model host–pathogen interactions in laboratory culture, with MDCK cells derived from canine kidney and A549 cells from human lung tissue. Researchers grow these cells as adherent monolayers, expose them to viruses or other infectious agents, and measure outcomes such as pathogen entry, replication, antigen expression, and cytopathic effects. Their distinct tissue origins provide complementary systems for studying epithelial barriers, innate immune responses, viral tropism, and infection dynamics. These models support pathogen characterization, antiviral screening, and interpretation of cellular mechanisms that influence respiratory and systemic infection.

We present a protocol describing a semi-quantitative method for measuring both, the attachment of influenza A virus to A549 cells, as well as the internalization of virus particles into the target cells by flow cytometry.

Generation of 3D Collagen-Embedded A549/MRC-5 Alveolar Organoids using a Mechanical Cell-Stretching Bioreactor

This protocol outlines the generation of organoids from A549 alveolar epithelial cells and MRC-5 lung fibroblasts on an extracellular matrix hydrogel scaffold, followed by embedding in 3D collagen gels and exposure to mechanical strain using a cell-stretching bioreactor to investigate alveolar mechanobiology.

Measuring Influenza Neutralizing Antibody Responses to A(H3N2) Viruses in Human Sera by Microneutralization Assays Using MDCK-SIAT1 Cells

Influenza neutralizing antibodies correlate with protection of influenza infections. Microneutralization assays measure neutralizing antibodies in human sera and are often used for influenza human serology. We describe a microneutralization assay using MDCK-SIAT1 cells to measure neutralizing antibody titers to contemporary 3C.2a and 3C.3a A(H3N2) viruses following influenza vaccination or infection.

Assessing Caspase-Mediated Cleavage of Viral and Host Proteins in Virus-Infected Cells

The video demonstrates a method to assess caspase-mediated protein cleavage in virus-infected cells. Post-viral infection, caspases get activated and degrade various host cellular proteins and viral nucleoproteins while specific caspase inhibitors prevent this degradation, as confirmed by western blotting.
