Sample Staining

Sample staining is a set of laboratory techniques that adds visible color or fluorescence to cells, tissues, or microorganisms, making their structures and molecular features easier to detect and compare. Stains work by selectively binding cellular components, microbial structures, or target molecules; preparation steps such as fixation and permeabilization help preserve samples and allow dyes or labeled antibodies to reach their targets. In immunology and infection research, staining supports the identification of pathogens, immune cell populations, tissue damage, and protein expression through light or fluorescence microscopy. These methods provide spatial context that complements culture, molecular assays, and flow cytometry in diagnosis and experimental investigation.

This protocol provides instruction for negative staining virus samples which can easily be used in BSL-2, -3, or -4 laboratories. It includes the use of an innovative processing capsule, which protects the transmission electron microscopy grid and provides the user easier handling in the more turbulent environments within biocontainment.

Here we describe a protocol for multiplex fluorescent immunohistochemical staining and imaging for the simultaneous localization of multiple cancer-associated antigens in lymphoma. This protocol can be extended to the colocalization analysis of biomarkers within all tissue sections.
