Ciprofloxacin Moxifloxacin

Ciprofloxacin and moxifloxacin are fluoroquinolone antibiotics that treat selected bacterial infections by targeting essential enzymes required for bacterial DNA replication. They bind to and inhibit DNA gyrase and, depending on the drug and organism, topoisomerase IV, causing DNA damage that prevents bacterial growth and survival. Their antibacterial spectra and tissue distribution differ, so clinicians select between them according to the suspected pathogen, infection site, and patient factors. Studying these medicines supports appropriate antimicrobial therapy while highlighting the importance of resistance surveillance and careful safety assessment, because fluoroquinolones can cause clinically significant adverse effects and should be used when benefits outweigh risks.

A High-throughput Compatible Assay to Evaluate Drug Efficacy against Macrophage Passaged Mycobacterium tuberculosis

New models and assays that would improve the early drug development process for next-generation anti-tuberculosis drugs are highly desirable. Here, we describe a quick, inexpensive, and BSL-2 compatible assay to evaluate drug efficacy against Mycobacterium tuberculosis that can be easily adapted for high-throughput screening.

This video demonstrates an assay to evaluate the efficacy of drugs against Mycobacterium tuberculosis (Mtb) infection. An in vitro infection model containing Mtb/macrophage aggregates is generated that mimics in vivo infection. Upon introducing suitable antibiotics, their efficacy is tested via resuzarin assay to monitor the survival of bacteria with increasing drug concentrations.

A Laser-induced Mouse Model of Chronic Ocular Hypertension to Characterize Visual Defects

Chronic ocular hypertension is induced using laser photocoagulation of the trabecular meshwork in mouse eyes. The intraocular pressure (IOP) is elevated for several months after laser treatment. The decrease of visual acuity and contrast sensitivity of experimental animals are monitored using the optomotor test.

Current in vitro models for evaluating contact lenses (CLs) and other eye-related applications are severely limited. The presented ocular platform simulates physiological tear flow, tear volume, air exposure and mechanical wear. This system is highly versatile and can be applied to various in vitro analyses with CLs.

Antimicrobial susceptibility testing of Mycobacterium tuberculosis is challenging but critical for patient care. This microtiter plate format offers testing of 12 antimycobacterial drugs and provides minimum inhibitory concentration (MIC) values, which will aid in appropriate treatment.
