Cytochrome P450 Induction

Cytochrome P450 induction is the process by which exposure to a chemical or drug increases the expression or activity of specific cytochrome P450 enzymes, altering how the body metabolizes substances. Inducing compounds often activate nuclear receptors such as the pregnane X receptor or constitutive androstane receptor, which regulate transcription of CYP genes and can accelerate hepatic drug metabolism. In medicine, this mechanism is central to understanding drug-drug interactions, as faster metabolism may reduce therapeutic drug concentrations or, for some prodrugs, increase formation of active metabolites. Studying induction supports safer prescribing, pharmacokinetic evaluation, dose adjustment, and prediction of treatment failure or toxicity.

Activation of Apoptosis by Cytoplasmic Microinjection of Cytochrome c

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.

This procedure describes how to encapsulate cytochrome c (cyt. c) in silica (SiO2) sol-gels, process these gels to form bioaerogels, and use these bioaerogels to rapidly recognize nitric oxide (NO) through a gas-phase reaction.
