Endothelial Junction Disruption

Endothelial junction disruption is the breakdown or loosening of contacts between vascular endothelial cells, a process that regulates passage of fluid, proteins, and leukocytes across the vessel wall. Inflammatory mediators, mechanical stress, or altered signaling can destabilize VE-cadherin and associated junctional proteins, while actomyosin contraction pulls neighboring cells apart and increases paracellular permeability. In medicine, this process helps explain tissue edema, vascular leak, and inflammatory injury, and it is studied in conditions such as sepsis, ischemia, and cancer. Measuring junctional integrity can support drug development aimed at preserving the endothelial barrier and controlling pathological vascular permeability.
