Hyperinsulinemic Clamp

The hyperinsulinemic clamp is a metabolic research method that measures how effectively the body responds to insulin, making it a reference technique for assessing insulin sensitivity. During the procedure, insulin is infused at a constant, controlled rate while glucose is infused and adjusted frequently to maintain stable blood glucose, usually at a euglycemic level. The glucose infusion rate required to preserve this target reflects the rate of glucose disposal and provides an estimate of whole-body insulin action. In medicine and metabolic research, the technique helps characterize insulin resistance, evaluate glucose regulation, and assess responses to treatments or physiological conditions.

Hyperinsulinemic-euglycemic Clamps in Conscious, Unrestrained Mice

The hyperinsulinemic-euglycemic clamp, or insulin clamp, is the gold standard for assessing insulin action in vivo. A method for performing insulin clamps in mice is described. This includes a method for arterial catheterization that permits experiments to be performed in conscious, unrestrained mice with minimal stress.

The hyperinsulinemic-euglycemic clamp is the "gold standard" for the assessment of insulin action. Insulin is infused at a constant rate stimulating glucose uptake. The amount of exogenous glucose infused to counter this drop is indicative of insulin sensitivity. Here the procedure is performed on a conscious, unrestrained rat.

Patch Clamp Electrophysiology

Neuron cell membranes are populated with ion channels that control the movement of charge into and out of the cell, thereby regulating neuron firing. One extremely useful technique for investigating the biophysical properties of these channels is called patch clamp recording. In this method, neuroscientists place a polished glass micropipette against a cell and apply suction to form a high resistance seal. This process isolates a small “patch” of membrane that contains one or more ion channels.

A hypoglycemic clamp is for measuring glucose production induced by counter-regulatory responses. Both methods use the same surgical procedure.
