Orthotopic Bladder Cancer

Orthotopic bladder cancer refers to bladder tumors that develop in, or are experimentally established within, the bladder’s native anatomical environment, making it a clinically relevant model for studying disease. In preclinical research, tumor cells or tissue are introduced into the bladder, where interactions with the urothelium, extracellular matrix, vasculature, and immune microenvironment can support tumor growth and invasion. These models help researchers examine cancer progression, local recurrence, metastasis, and responses to chemotherapy, immunotherapy, or intravesical treatments. By more closely reproducing tumor location and microenvironment than ectopic models, orthotopic bladder cancer research can improve therapeutic evaluation and translational medicine.

Implantation of cancer cells into the organ of origin can serve as a useful preclinical model to evaluate novel therapies. MB49 bladder carcinoma cells can be grown within the bladder following intravesical instillation. This protocol demonstrates catheterization of the mouse bladder for the purpose of tumor implantation and adenoviral delivery.
