Hamburger Hamilton

The Hamburger-Hamilton system is a standardized morphological staging scale that describes developmental progress in chick embryos, providing a common reference for embryology and neuroscience research. Rather than relying only on incubation time, it assigns embryos to numbered stages using visible landmarks such as body size, somite formation, limb development, eye and brain morphology, and axial flexure. In neuroscience, this framework allows researchers to align experiments with comparable phases of neural tube, brain, and spinal cord development, improving reproducibility when analyzing patterning, neuronal differentiation, and developmental defects. Its shared staging language also supports precise sampling, imaging, and cross-study comparisons.

A method by which gene expression in the neural tube can be downregulated in a cell type-specific, traceable manner is described. We demonstrate how in ovo electroporation of microRNA-based plasmids that elicit spatiotemporally controlled RNA interference can be used to investigate commissural axon guidance in the developing neural tube.

Ectopic expression is one technique to elucidate the microRNAs role in brain development. However, targeting specific areas using in ovo electroporation is challenging. Here, we show an efficient way to selectively electroporate ventral and dorsal midbrain regions.
