Store Operated Calcium Entry

Store-operated calcium entry (SOCE) is a cellular mechanism that replenishes cytoplasmic calcium when calcium stores in the endoplasmic reticulum become depleted, supporting signaling in neurons and other cells. Depletion activates the sensor STIM1, which undergoes conformational changes and clusters near the plasma membrane to open Orai1 calcium channels, allowing extracellular calcium to enter the cell. In neuroscience, SOCE helps regulate neuronal excitability, gene expression, synaptic function, and calcium-dependent survival pathways. Studying this process clarifies how neurons maintain calcium balance and may reveal mechanisms involved in neurodevelopmental disorders, neurodegeneration, and altered circuit function.

The extent of store-operated Ca2+ entry (SOCE) can be monitored using fluorescent Ca2+ indicators. Mn2+ quenching of such indicators assays SOCE in cultured cells and skeletal muscle fibers. A technique allowing spatial and temporal resolution of SOCE by confocal imaging of mechanically skinned muscle fibers is also described.

Energy Stored in a Capacitor

2023

When an archer pulls the string in a bow, he saves the work done in the form of elastic potential energy. When he releases the string, the potential energy is released as kinetic energy of the arrow. A capacitor works on the same principle in which the work done is saved as electric potential energy. The potential energy (UC) could be calculated by measuring the work done (W) to charge the capacitor. Let us consider the case of a parallel plate capacitor. When the capacitor is connected to a...
