This article demonstrates a protocol for quantifying bacterial load in spleen and liver tissues from mice experimentally infected with an intracellular bacterial pathogen, such as Listeria monocytogenes. The method involves tissue collection, mechanical and detergent-based lysis, serial dilution, plating on nutrient agar, and colony counting to assess bacterial burden in each organ.
Quantitative measurement of bacterial load in mouse spleen and liver provides a robust readout for evaluating host-pathogen interactions and immune response efficacy in preclinical infectious disease models. This approach enables mechanistic de-risking and supports predictive confidence in target validation for anti-infective drug discovery. Accurate organ-specific bacterial quantification informs portfolio decisions at key discovery and lead identification inflection points.
This quantification method integrates into the discovery-to-preclinical continuum, supporting both target validation and lead identification in infectious disease research.
