Fluorescence Leakage Assay

A fluorescence leakage assay is a biochemical method for measuring membrane permeability by tracking the release of a fluorescent marker from lipid vesicles or other membrane-bound compartments. The assay typically encapsulates a dye at a self-quenching concentration; when a membrane-disrupting agent forms pores or otherwise compromises the bilayer, dye escapes, becomes diluted, and produces an increase in fluorescence. Researchers use the resulting signal to quantify membrane damage and compare the effects of antimicrobial peptides, proteins, detergents, drugs, or other compounds on lipid organization. This approach provides a sensitive, real-time readout of membrane stability and molecular interactions.
