Fragment Libraries

Fragment libraries are collections of small, low-molecular-weight chemical compounds used to identify starting points for ligand discovery and drug design. In biochemistry, each fragment typically binds weakly to a target, and screening methods such as X-ray crystallography, nuclear magnetic resonance, or biophysical assays detect and characterize these interactions. Because fragments sample chemical space efficiently, researchers can optimize confirmed hits by adding groups that improve affinity, selectivity, and physicochemical properties while preserving the binding mode. Fragment-based discovery therefore supports structure-guided development of enzyme inhibitors and other bioactive molecules, often providing novel scaffolds when conventional screening yields few suitable leads.

Bacterial Inner-membrane Display for Screening a Library of Antibody Fragments

We provide a method to simultaneously screen a library of antibody fragments for binding affinity and cytoplasmic solubility by using the Escherichia coli twin-arginine translocation pathway, which has an inherent quality control mechanism for intracellular protein folding, to display the antibody fragments on the inner membrane.

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.

A Universal Protocol for Large-scale gRNA Library Production from any DNA Source

Methods for generating large-scale gRNA libraries should be simple, efficient and cost-effective. We describe a protocol for the production of gRNA libraries based on enzymatic digestion of target DNA. This method, CORALINA (comprehensive gRNA library generation through controlled nuclease activity) presents an alternative to costly custom oligonucleotide synthesis.
