Glycerol Gradient Fractionation

Glycerol gradient fractionation is a biochemical separation technique that resolves proteins, protein complexes, nucleic acids, or organelles according to their sedimentation behavior in a glycerol density gradient. During centrifugation, particles move through progressively denser glycerol layers at rates determined primarily by size, shape, and mass, while the gradient reduces mixing and helps preserve labile complexes. Researchers collect sequential fractions from the gradient and analyze them using methods such as enzymatic assays, electrophoresis, or immunoblotting. This approach supports the study of macromolecular assembly, oligomerization, molecular weight estimates, enzyme complexes, and interactions that are difficult to characterize in unfractionated samples.
