Hyperlipidemia Models

Hyperlipidemia models are experimental systems that reproduce elevated levels of lipids or lipoproteins in living organisms or cultured cells, enabling controlled investigation of lipid metabolism and its biochemical consequences. Researchers create these models through dietary manipulation, genetic alteration, or pharmacological intervention, then measure changes in cholesterol, triglycerides, lipoprotein transport, and related metabolic pathways. In biochemistry, hyperlipidemia models help clarify how lipid synthesis, uptake, storage, and clearance become disrupted. They also support evaluation of disease mechanisms, comparison of therapeutic strategies, and assessment of candidate drugs aimed at restoring lipid balance and reducing hyperlipidemia-associated complications.

This protocol presents a detailed approach to replicating a murine model of hyperlipidemia-induced heart failure with preserved ejection fraction (HFpEF). The design combines the administration of adeno-associated virus 9-cardiac troponin T-low-density lipoprotein receptor (AAV9-cTnT-LDLR) and poloxamer-407 (P-407).

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.
