Maldi Sample Preparation

MALDI sample preparation is the process of combining an analyte with a suitable matrix to enable matrix-assisted laser desorption/ionization mass spectrometry, a technique used to measure biomolecules by their mass-to-charge ratios. Typically, the sample is mixed with an ultraviolet-absorbing organic matrix and dried on a target plate, where co-crystallization allows the matrix to absorb laser energy, desorb the analyte, and promote ion formation with limited fragmentation. Careful control of solvent composition, analyte concentration, matrix selection, and crystallization improves signal quality and reproducibility. In biochemistry, MALDI sample preparation supports the analysis of peptides, proteins, lipids, carbohydrates, and microbial biomolecules for identification and structural characterization.
