Plant Inoculation

Plant inoculation is the deliberate introduction of microorganisms or other biological agents into a plant, its seeds, or surrounding soil to establish a controlled biological interaction. In bioengineering, inoculants such as rhizobacteria, fungi, or engineered strains are applied to specific plant sites, where they colonize tissues or the rhizosphere and influence nutrient exchange, signaling, growth, or defense responses. Researchers use plant inoculation to study plant–microbe relationships and develop biofertilizers, biocontrol strategies, and engineered systems that improve nutrient acquisition, stress tolerance, or disease resistance. Controlled inoculation also supports reproducible testing of microbial functions under defined environmental conditions.

Here, we present a protocol to test plant virulence with the plant pathogen Magnaporthe grisea. This report will contribute to the large-scale screening of the pathotypes of fungi isolates and serve as an excellent starting point for understanding the resistant mechanisms of plants during molecular breeding.

A Spray Inoculation Technique to Establish Fungal Infection in Rice Plant Leaves

Improving our understanding of and our ability to manage many of the insect-transmitted plant viruses requires the use of the vector. Insect transmission of plant viruses is a tritrophic interaction, and as such requires the manipulation of insects, virus, and plant. Vectors must be reared in large numbers and manipulated in such a way as to insure high rates of transmission to test plants. The basics of rearing and manipulating the whitefly, Bemisa tabaci (Hemiptera: Aleyrodidae), a vector of...

Establishment of Rice Plant Leaf Infection through Fungal Inoculation via Wounding

2025

This video demonstrates the establishment of rice plant leaf infection through wounding inoculation with Magnaporthe grisea mycelium. The fungal spore-derived germ tube penetrates the epidermal layer, resulting in tissue infection and lesions on the leaves. This method aids pathotype screening and improves plant resistance understanding for molecular breeding.
