Chambered Slide Imaging

Chambered slide imaging is a microscopy technique that captures biological samples within individual chambers on a slide, enabling organized observation of cells, tissues, or microorganisms under laboratory conditions. The chambers confine the specimen and retain culture medium or staining reagents while a microscope records transmitted-light or fluorescence images, including time-lapse changes in living samples. In biology, this approach supports analysis of cell morphology, growth, viability, migration, and fluorescent marker distribution. Its slide-based format allows multiple conditions or samples to be examined in parallel, providing reproducible visual data for cell biology, developmental studies, disease research, and experimental screening.

The Drosophila egg chamber is an excellent model for studying the mechanisms of mRNA localization. In order to capture the dynamic events that underpin the processes of localization, rapid high resolution imaging of live tissue is required. Here, we present a protocol for dissection and imaging of live samples with minimal disruption.
