Egg Hatching Assay

An egg hatching assay is a biological test that measures whether and when eggs hatch under defined experimental conditions, providing a practical endpoint for development, viability, or biological activity. In the assay, eggs are maintained in a controlled environment and exposed to a test substance or condition; investigators monitor embryo development and record the proportion or timing of hatching, typically relative to an untreated control. This approach supports studies of embryology, environmental effects, and compound screening by revealing developmental inhibition or altered hatch rates. Standardized conditions and consistent scoring improve reproducibility and help distinguish treatment effects from natural variation.

Embryonic stages are the most susceptible to xenobiotics. Although chemical toxicity depends on salinity, no method exists to test the salinity dependence of toxicity to aquatic organisms. Here, we describe a new and high-throughput method for determining the salinity dependence of toxicity to aquatic embryos.

2023

Drosophila females choose between substrates and select sites that are attractive for egg-laying. This video describes a behavioral assay that tests their decision-making, called the egg-laying preference assay. The featured clip demonstrates the procedure with a custom-built setup, which enables researchers not only to test the behavior of many females at the same time but is also compatible with video recording and optogenetic setups.
