Gfap Reporter Construct

A GFAP reporter construct is a genetic tool that links regulatory sequences from the glial fibrillary acidic protein (GFAP) gene to a detectable reporter, such as fluorescent protein, to monitor GFAP-associated gene activity. In cells carrying the construct, GFAP promoter or enhancer elements drive reporter expression when the relevant transcriptional programs are active, providing a visual or measurable readout of astrocyte-lineage states and related changes. In cancer research, these constructs help identify and track glial tumor cells, assess cellular heterogeneity and differentiation, and examine how signaling pathways or treatments alter tumor-associated phenotypes. They can also support imaging, cell sorting, and functional studies.

MicroRNAs (miRNAs) are important regulators of gene expression and have been shown to play a role in numerous biological processes. To better understand miRNA-UTR interactions, we have created a genome-wide collection of 3 UTR luciferase reporters paired with a novel luciferase gene and assay reagent, the LightSwitch system.

2025

This video demonstrates a technique to generate a glioblastoma stem cell reporter line to identify the differentiation of glioblastoma stem cells (GSCs) into astroglia. It outlines the steps involved in seeding GSCs, transducing them with a lentivirus encoding GFP, and determining GFP reporter expression through flow cytometry.

Construction of CRISPR Plasmids and Detection of Knockout Efficiency in Mammalian Cells through a Dual Luciferase Reporter System

2020

Here, we present a protocol describing a streamlined method for the efficient generation of plasmids expressing both the CRISPR enzyme and associated single guide RNA (sgRNAs). Co-transfection of mammalian cells with this sgRNA/CRISPR vector and a dual luciferase reporter vector that examines double-strand break repair allows evaluation of knockout efficiency.

Data Reporting and Recording

Reporting and recording are crucial in data documentation. The timely, thorough, and accurate documentation of facts is essential when recording patient data. Failure to record findings during an assessment or interpretation of a problem will result in loss of information and make the patient document unreliable. The reader is left with general impressions if the information is not specific. A recording is documenting data of the individual's health information in a traceable, secure, and...
