Chemoattractant Treatment

Chemoattractant treatment is the controlled exposure of cells to chemical cues that direct their movement along a concentration gradient, a process known as chemotaxis. Chemoattractants such as chemokines or other signaling molecules bind receptors on the cell surface, activating intracellular pathways that reorganize the cytoskeleton and drive migration toward higher or lower concentrations. In cancer research, this approach helps measure how tumor cells, immune cells, and stromal cells respond to signals within the tumor microenvironment. These assays clarify mechanisms of invasion, metastasis, immune-cell recruitment, and cell trafficking, supporting the evaluation of therapeutic strategies that disrupt cancer-promoting migration.

We describe a method to construct devices for 3D culture and experimentation with cells and multicellular organoids. This device allows analysis of cellular responses to soluble signals in 3D microenvironments with defined chemoattractant gradients. Organoids are better than single cells at detection of weak noisy inputs.

Using frog sperm we describe and compare two classes of assays used to detect sperm chemoattraction – sperm accumulation assays and sperm tracking assays.
