Glucose Transporter 1 Expression

Glucose Transporter 1 (GLUT1) expression refers to the production and abundance of GLUT1, a membrane protein that enables many cells to take up glucose. GLUT1 transports glucose across the plasma membrane by facilitated diffusion, moving it down its concentration gradient without directly using ATP. In cancer research, elevated GLUT1 expression is commonly examined as a sign of increased glucose demand and altered metabolism, particularly in rapidly growing or oxygen-limited tumors. Measuring GLUT1 through gene expression, protein assays, or tissue imaging can help characterize tumor biology, evaluate metabolic adaptations, and support research into diagnostic markers and metabolism-targeted therapies.

Monocytes are integral components of the human innate immune system that rely on glycolytic metabolism when activated. We describe a flow cytometry protocol to measure glucose transporter expression and glucose uptake by total monocytes and monocyte subpopulations in fresh whole blood.

This manuscript describes how to screen for thermostabilizing mutations, purify the human serotonin transporter, generate high affinity antibodies, and crystallize the serotonin transporter-antibody complex bound to the antidepressant drug S-citalopram. This protocol can be adapted to the study of other challenging membrane transporters, receptors, and channels.

We describe simple methods to study the regulation of intestinal serotonin transporter (SERT) function and expression using an in vitro cell culture model of Caco-2 cells grown in 3D and an ex vivo model of mouse intestines. These methods are applicable to the study of other epithelial transporters.
