Osmium Tetroxide Post-fixation

Osmium tetroxide post-fixation is a secondary fixation step used after aldehyde fixation to preserve and visualize cellular ultrastructure, particularly in transmission electron microscopy. Osmium tetroxide reacts with unsaturated lipids, stabilizing biological membranes and adding electron density before dehydration, resin embedding, and ultrathin sectioning; this improves contrast of organelles, membrane junctions, and lipid-rich structures. In cancer research, the method supports detailed examination of tumor-cell morphology, mitochondrial and membrane alterations, cell-cell contacts, and interactions with the surrounding microenvironment, helping researchers connect structural changes with malignant behavior, treatment response, and disease progression.

A novel non-cross-linking, formalin-free tissue fixative that allows high quality morphologic, molecular and FISH analyses from the same specimen by addition of a post-fixation step before FISH is presented.
