Branched Gdgts

Branched GDGTs (glycerol dialkyl glycerol tetraethers) are membrane lipids produced primarily by soil and sediment bacteria, and their molecular distributions preserve information about past environmental conditions. Their structures contain branched carbon chains whose numbers of methyl groups and cyclopentane rings vary with growth temperature and, for some compounds, soil or sediment pH. Researchers measure these lipid patterns using chromatographic and mass-spectrometric methods to calculate environmental proxies such as the methylation and cyclization of branched tetraethers. In environmental sciences, branched GDGTs help reconstruct terrestrial temperature, hydroclimate, soil development, and ecosystem responses to environmental change.

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.
