Comparative Ct Method

The Comparative Ct Method, also called the 2−ΔΔCt method, is a quantitative real-time PCR approach used to estimate relative changes in gene expression between samples. It compares the cycle threshold (Ct) of a target gene with that of a stable reference gene, then contrasts the normalized target value in an experimental sample with a calibrator sample; assuming comparable amplification efficiencies, the resulting 2−ΔΔCt value represents fold change. In genetics, this method supports analysis of transcriptional responses, allele- or condition-associated expression differences, and molecular effects of mutations or treatments. Its relative design provides a practical alternative to absolute quantification when an appropriate reference and calibrator are available.
