Propidium Iodide Ribonuclease

Propidium iodide ribonuclease staining is a fluorescence-based method for measuring cellular DNA content, making it useful for analyzing genome replication and cell-cycle distribution in genetics research. After cells are fixed and permeabilized, ribonuclease digests RNA, while propidium iodide intercalates into the remaining DNA and emits fluorescence proportional to its abundance. Flow cytometry then quantifies fluorescence from individual cells, distinguishing DNA-content populations associated with G0/G1, S, and G2/M phases and identifying abnormal ploidy or sub-G1 DNA. The method supports studies of cell proliferation, chromosome content, cell-cycle regulation, and responses to genetic or chemical perturbations.
