Antigenic Challenge

Antigenic challenge is the exposure of an immune system to a foreign antigen, providing a stimulus that reveals how effectively it recognizes and responds to potential infection. Antigen-presenting cells process the antigen and display fragments on major histocompatibility complex molecules, activating antigen-specific T cells and supporting B-cell proliferation, antibody production, and immune memory. In immunology and infection research, controlled antigenic challenges help characterize host defenses, compare primary and secondary responses, and evaluate vaccine-induced protection. Measuring cellular responses, antibody titers, and pathogen control after challenge can clarify mechanisms of immunity and guide the development of vaccines and other interventions.

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).
