Hepatic Artery Injection

Hepatic artery injection is a technique for delivering cells, pathogens, drugs, or other test materials directly into the liver’s arterial blood supply, making it useful for studying hepatic biology and immune responses. A needle or catheter places the material in the hepatic artery, where blood flow distributes it through branching vessels and exposes liver tissue, including hepatocytes and resident immune cells. In immunology and infection research, this approach can help model localized exposure, examine host–pathogen interactions, evaluate immune activation, and assess tissue-specific treatment effects. Controlled delivery also supports comparisons of dose, distribution, and inflammatory outcomes in experimental systems.

Targeting Liver Tumors with Oncolytic Viruses via the Hepatic Artery

The video demonstrates locoregional delivery of oncolytic viruses through hepatic arterial injection as a therapeutic approach for hepatocellular carcinoma. These viruses selectively replicate in tumor cells, inducing cell lysis and releasing tumor cell-associated antigens. This process triggers immunogenic cell death, inducing an anti-tumor response.

Auxiliary liver transplantation provides a temporary support in acute hepatic failure, until regeneration of the failing liver. The heterotopic auxiliary liver transplantation (HALT) with portal vein arterialization (PVA) renders sufficient liver function. We developed an analogous technique in the rat, to examine the influence of the portal vein arterialization on the morphology and function of the graft.

This manuscript provides the detailed procedure of intra-iliac artery (IIA) injection, a technique to deliver cancer cells specifically to hind limb tissues including bones to establish experimental bone metastases. Although initially established with breast tumor models, this protocol can be easily extended to other cancer types.
