Mrna Vaccine

An mRNA vaccine is an immunization that delivers messenger RNA instructions to cells so they can produce a harmless piece of a pathogen, preparing the immune system to recognize it. Typically enclosed in lipid nanoparticles, the mRNA enters cells and is translated into an antigen, which activates innate and adaptive immune responses, including antibody production and T-cell activity; the mRNA is then broken down. In immunology and infection research, this platform supports rapid vaccine design against emerging pathogens and enables targeted evaluation of antigen selection, immune protection, and vaccine effectiveness. Its flexible manufacturing approach also supports continued development of vaccines for infectious diseases.

Here, a simple protocol is presented for producing mRNA nanoparticles based on poly(beta aminoester) polymers, easy to be tailored by changing the encapsulated mRNA. The workflow for synthesizing the polymers, the nanoparticles, and their in vitro essential characterization are also described. A proof-of-concept regarding immunization is also added.
