Tnf Alpha Neutralization

TNF alpha neutralization is the targeted reduction or blockade of tumor necrosis factor alpha, a proinflammatory cytokine that coordinates immune-cell activation and inflammatory responses. It works when monoclonal antibodies or soluble receptor-based agents bind TNF alpha, preventing it from engaging cell-surface TNF receptors and triggering downstream signaling. In immunology and infection research, this approach helps clarify TNF alpha’s roles in fever, leukocyte recruitment, tissue inflammation, and granuloma maintenance, while supporting studies of anti-inflammatory therapies for autoimmune disease. Because TNF alpha also contributes to host defense, neutralization can weaken containment of certain infections, making infection risk and immune monitoring important considerations.

Optimization of a Quantitative Micro-neutralization Assay

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.
