Matrigel Cell Suspension

Matrigel cell suspension is a three-dimensional culture preparation in which cells are mixed with or embedded in Matrigel, a basement membrane matrix used to model tissue-like environments in vitro. Kept cold, Matrigel remains fluid for cell handling; at physiological temperature, its extracellular matrix proteins, including laminin and collagen IV, form a gel that supports cell attachment, organization, and growth. In medicine and biomedical research, this system enables organoid formation, cancer modeling, stem cell studies, and drug response testing. By providing structural and biochemical cues that conventional two-dimensional cultures lack, Matrigel cell suspensions help researchers investigate cell behavior and evaluate potential therapies.

This video demonstrates how to maintain the growth of human embryonic stem cells (hESCs) in feeder cell-free conditions and how to continuously passage hESCs in feeder cell-free conditions. Confirmation of hESC pluripotency grown in feeder cell-free conditions by immunofluorescence microscopy is also demonstrated. Part 3 of 3.

Dissociating cells from specific tissue types requires specific parameters for tissue aggitation to obtain a high volume of viable, culturable cells. The Miltenyi gentleMACS Dissociator optimizes this task with a simple, practical protocol. In this publication the use of this apparatus on nerual tissue is explained.
