Mouse Aortic Graft

A mouse aortic graft is an experimental vascular transplantation model in which a segment of aorta is surgically transferred between mice to study blood vessel biology and graft responses. Researchers connect the donor vessel to the recipient circulation through precise microvascular anastomoses, allowing blood flow to expose the graft to hemodynamic forces, immune surveillance, and tissue remodeling. This model supports investigation of transplant rejection, vascular inflammation, atherosclerosis, aneurysm development, and graft healing under controlled genetic and treatment conditions. Because researchers can monitor structural and molecular changes over time, mouse aortic grafts help evaluate disease mechanisms and potential therapies for vascular and transplant medicine.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.

To improve our knowledge of cellular and molecular neotissue formation, a murine model of the TEVG was recently developed. The grafts were implanted as infrarenal vena cava interposition grafts in C57BL/6 mice. This model achieves similar results to those achieved in our clinical investigation, but over a far shortened time-course.
