Type 2 Inflammation

Type 2 inflammation is an immune response that protects against parasites and supports tissue repair but can drive allergic and other chronic diseases when excessive or persistent. It is coordinated by signals such as interleukin-4, interleukin-5, and interleukin-13, produced by T helper 2 cells and innate lymphoid cells, which promote immunoglobulin E production, eosinophil recruitment, mucus secretion, and changes in epithelial barrier function. In medicine, identifying type 2 inflammation helps explain conditions including asthma, atopic dermatitis, and chronic rhinosinusitis, while guiding biomarker-based diagnosis and targeted treatment with biologic therapies that block specific cytokine pathways.

Overview In response to tissue injury and infection, mast cells initiate inflammation. Mast cells release chemicals that increase the permeability of adjacent blood capillaries and attract additional immune cells to the wound or site of infection. Neutrophils are phagocytic leukocytes that exit the bloodstream and engulf invading microbes. Blood clotting platelets seal the wound and fibers create a scaffold for wound healing. Macrophages engulf aging neutrophils to end the acute inflammatory...

Diabetes Mellitus: Type 2 and Gestational

2024

Type 2 diabetes, characterized by insulin resistance, arises when the insulin receptors on cells lose responsiveness to insulin, diminishing the cell's capacity to take up glucose, resulting in elevated blood glucose levels. To receive a diagnosis of Type 2 diabetes, a series of blood glucose tests are necessary to assess whether the blood glucose falls within normal parameters. If the result is out of the normal range, a patient may be diagnosed as prediabetic or diabetic, depending on the...

This protocol provides a simple method of making static training equipment for mice. The device maintains the muscle isometric contraction of the limbs of mice so as to verify the intervention effect of traditional exercise on type 2 diabetes (T2DM) and provides new exercise therapy for the clinical treatment of T2DM.

Here we describe a novel high-content chemically induced inflammation assay aiming at the identification of immune-modulatory bioactives. We have successfully combined automated microscopy with custom developed software scripts enabling automated quantification of the inflammatory response as well as further data processing, analysis, mining, and storage.
