Vascularization Dynamics

Vascularization dynamics describes how blood vessel networks form, remodel, and regulate blood flow over time in response to changing tissue demands. This process involves endothelial cell activation, migration, and proliferation, followed by vessel maturation, branching, regression, and adjustment of perfusion through signals such as oxygen availability and growth factors. In medicine, understanding these dynamics helps explain wound healing, tumor progression, ischemic disease, and the development of engineered tissues. Measuring vascular growth and function can support disease modeling, therapeutic design, and strategies for improving tissue repair and graft integration.

Intravital Longitudinal Imaging of Vascular Dynamics in the Calvarial Bone Marrow

Intravital microscopy allows the study of dynamic biological processes such as tissue regeneration and tumor development. The calvarial bone marrow, a highly dynamic tissue, offers insights into hematopoiesis and vascular function. Using a biocompatible 3D-printed head fixation implant allows for repetitive longitudinal imaging, enhancing our understanding of tissue dynamics and the tumor microenvironment.

To date, thick tissue defects are typically reconstructed by applying autologous tissue flaps or engineered tissues. In this protocol, we present a new method for engineering vascularized tissue flap bearing an autologous pedicle, to serve as a substitute to autologous flaps.

Tools used for visualizing vascular regeneration require methods for contrasting the vascular trees. This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.
