Ampa Nmda Ratio

The AMPA/NMDA ratio is an electrophysiological measure comparing currents mediated by AMPA-type and NMDA-type glutamate receptors at excitatory synapses, helping characterize synaptic strength and development. Researchers typically record synaptic responses under voltage-clamp conditions, measuring the rapid AMPA receptor current at negative holding potentials and the slower NMDA receptor current at depolarized potentials after AMPA-mediated responses decline. Changes in this ratio can indicate synaptic maturation, receptor trafficking, or long-term potentiation and depression. In neuroscience, the measure supports studies of learning, memory, neural development, and disease-related alterations in glutamatergic transmission.

Here, we describe the use of a genetically directed alkaline phosphatase (AP) labeling method to investigate the anatomical features and phenotypes of mouse retinas under NMDA-induced excitotoxicity. In conjunction with several retinal ganglion cell (RGC) subtype-specific reporter lines, we uncover novel phenotypes in dying RGCs throughout the degenerative process.
