Constitutive Expression

Constitutive expression is the continuous production of a gene product under conditions in which its regulatory sequence remains active, rather than being switched on by a specific stimulus. In engineered cells, this typically results from placing a gene under a constitutive promoter, allowing transcription and subsequent translation to proceed with relatively stable activity over time, although expression levels can vary with cell state and construct design. In neuroscience, constitutive expression supports persistent production of fluorescent reporters, receptors, ion-channel proteins, or other molecular tools in cultured neurons and animal models. This approach enables cell labeling, circuit tracing, and long-term analysis of neuronal structure and function, while requiring attention to dosage and potential effects on cell physiology.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
